In the realm of suspension cell culture, traditional fed-batch bioreactor systems, which usually run between 7–21 days, have long been the standard. These systems involve cells consuming media nutrients and releasing both toxic metabolites and the targeted protein into the media. At the end of the run, the protein is then separated from the cell mass. Even with numerous technological enhancements, fed-batch processes still grapple with issues such as large manufacturing footprints and scalability challenges.

In contrast, perfusion bioreactors, a pivotal element in cutting-edge culture technologies, offer extended cell culture periods, even spanning several months. This is achieved by continuously feeding the cells with fresh media and concurrently removing the spent media. 

When it comes to separating cells from the culture fluid during spent media removal, several methods are employed. The tangential flow filtration system (TFF) and its counterpart, the alternating tangential filtration system (ATF), are predominant. They maintain cells within the bioreactor while facilitating media removal. Centrifugation offers an alternative approach, isolating cells and subsequently reintroducing them into the bioreactor.

However, there exist limitations with current perfusion system bioreactor separation devices. Issues such as limited perfusion batch scalability (maxing out at 1000L), frequent filter blockages, and the exertion of shearing forces on cells during separation present significant challenges.

As a result, the bioprocess industry still awaits the development of both perfusion-enabled micro bioreactors (100 ml) and upscaled versions of perfusion bioreactors exceeding 2000L.
